ABSTRACT

Este estudo teve como objetivo caracterizar a epidemiologia da presença de ácaros hematófagos em granjas de postura no estado de Minas Gerais. Foi utilizado um banco de dados secundário, com informações de 402 galpões de 42 propriedades comerciais. As variáveis utilizadas para compor o modelo de correspondência foram selecionadas por meio do teste qui-quadrado (P≤0,05). Foi construído um índice para a presença de ácaros hematófagos, considerando alguns fatores de risco. Além disso, um estudo da análise espacial foi realizado para avaliar a presença de ácaros hematófagos em Minas Gerais. Observou-se a presença de ácaros hematófagos em 48% dos galpões, sendo a de O. sylviarum de 45,5%, O. bursa de 17,4% e D. gallinae de 2,7%. Houve associação entre o índice de risco com os intervalos de remoção de fezes e com a presença ou a ausência de aves sinantrópicas. Verificou-se que granjas mais tecnificadas não utilizam acaricida em seus galpões. A presença de ácaros hematófagos foi observada na mesorregião Sul/Sudoeste de Minas Gerais e entre as mesorregiões Oeste de Minas e Metropolitana de Belo Horizonte. Esses resultados fornecem conhecimento sobre a epidemiologia desses ectoparasitos e podem contribuir na tomada de decisões, reduzindo os riscos de possíveis infestações em aves de postura.(AU)


This study aimed to characterize the epidemiology of the presence of hematophagous mites in posture farms in the state of Minas Gerais. Was used a secondary database with information from 402 chicken houses in 42 commercial properties. The variables used to compose the correspondence model were selected through chi-square tests (P≤0.05). A risk index was built to the presence of hematophagous mites considering some risk factors. Additionally, a study of the spatial analysis was done to evaluate the presence of hematophagous mites in Minas Gerais. Was observed the presence of hematophagous mites in 48% of chicken houses, the presence of O. sylviarum was 45.5%, O. bursa 17.4% and D. gallinae 2.7%. There was an association between the risk index with the intervals for the removal of manure and the presence or absence of synanthropic birds. It was verified that more technified farms do not use acaricide in their chicken houses. The presence of hematophagous mites was observed in the Messoregions South/Southwest of Minas Gerais and between the West and Metropolitan Mesoregion of Belo Horizonte. These results provide knowledge about the epidemiology of these ectoparasites and may contribute to decision making by reducing the risks of possible infestations in poultry.(AU)

ABSTRACT

This study aimed to evaluate the neutrophil oxidative metabolism and phagocytosis of Zymonsan particles of primiparous and pluriparous Lacaune ewes during the first 30 days after lambing. A total of 20 ewes were evaluated, 10 primiparous (GPR) and 10 pluriparous (GPL). Evaluation of basal oxidative metabolism was performed using the nitroblue tetrazolium (NBT) technique, stimulating neutrophil phagocytosis with Zymosan particles. Blood samples were collected at parturition day (M1) and 1, 3, 7, 15 and 30 days after parturition, corresponding to M2, M3, M4, M5 and M6, respectively. In relation to the groups, GPR presented lower oxidative basal metabolism neutrophils in M1 compared to M4, in M3 with M1, M2, M4 and M5. In M4 and M5 differences were found at all times and in M6 with M4 and M5. Higher percentage of neutrophils than phagocytes were found in M4, M5 and M6 than in M1 and M2 in GPL animals. At all times GPR presented a lower percentage of phagocytosis than GPL. Thus it is concluded that the immune response in pluriparous sheep was more efficient than in primiparous sheep.(AU)

ABSTRACT

A total of 480 milk samples were analyzed in four repetitions with four preservative treatments (no preservative, Bronopol, Bronolat and Brononata), three storage times at temperatures up to 4 °C (24, 48 and 72hours after reception), five different water additions (0.0, 2.5, 5.0, 7.5 and 10.0%) and two analytical instruments (electronic cryoscope and FTIR). The objective of this study was to evaluate the effect of these parameters in the determination of the freezing point by the reference method and by Fourier transform infrared spectroscopy, thus determining the best analytical conditions and establishing a mathematical equation for electronic determination by FTIR spectroscopy. Bronolat was the best preservative and Brononata was the worst and is not recommended to analyze freezing point by FTIR. The storage time of the samples did not interfere in the analytical determinations by the precision method and by FTIR.(AU)


Foram analisadas 480 amostras de leite em quatro repetições em relação a quatro tratamentos por conservantes (sem conservante, bronopol, bronolat e brononata), três tempos de armazenamento, em temperatura até 4ºC desde a recepção da amostra (24, 48 e 72 horas), cinco porcentagens de adição de água (0,0; 2,5; 5,0; 7,5 e 10,0%) e dois instrumentos analíticos (crioscópio eletrônico e FTIR). O objetivo foi avaliar o efeito desses parâmetros na determinação do índice crioscópico pelo método de precisão em crioscópio eletrônico e por espectroscopia com transformada de Fourier no infravermelho, determinando-se, assim, as melhores condições analíticas. Entre os conservantes utilizados, bronolat foi o melhor e brononata foi o menos eficiente, não sendo, portanto, recomendado para análise de crioscopia por FTIR. O tempo de armazenamento das amostras não interferiu nas determinações analíticas pelo método de precisão e por FTIR.(AU)

ABSTRACT

INTRODUCTION: Mononeuritis multiplex (MM) is an unusual form of peripheral neuropathy involving at least two noncontiguous peripheral nerve trunks. The pure sensory form of MM occurs rarely. Immunoglobulin (Ig)G subclass deficiency is a clinically and genetically heterogeneous disorder. Up to 50% of adults with selective subnormal IgG1 levels or selective IgG1 deficiency have a concomitant autoimmune disorder. Herein, we report the case of a patient with MM and selective IgG1 deficiency who showed remarkable clinical improvement after 2-year combination therapy with the DPP-4 inhibitor sitagliptin plus vitamin D3. CASE REPORT: A 49-year-old man developed numbness in right hand and forearm. After 6 months, the patient developed left forefoot numbness. Approximately 8 years later, the patient started to develop numbness also in the right forefoot, along with symptoms of evening fatigue and occasional orthostatic hypotension. The patient also reported recurrent candidiasis in glans and intergluteal areas since adolescence. Electromyoneurography of lower and upper limbs revealed the presence of multiple mononeuropathies. Protein electrophoresis showed hypogammaglobulinemia and low serum IgG1 levels. Sural nerve biopsy showed the presence of perineuritis. The patient was diagnosed with MM due to perineuritis probably secondary to IgG1 deficiency. We, then, proposed combination therapy with sitagliptin and vitamin D3 in the attempt to achieve immunomodulation. At the last follow-up visit (2 years), the patient showed persistent clinical improvement, increase in IgG1 levels and normalization of protein electrophoresis. CONCLUSIONS: To the best of our knowledge, this is the first case showing a remarkable clinical improvement of MM and selective IgG1 deficiency achieved through a combination therapy with sitagliptin and vitamin D3.

ABSTRACT

Objetivou-se conhecer o perfil epidemiológico do paciente que buscou atendimento antirrábico pós-exposição associado a acidentes com gatos domésticos, em Belo Horizonte/MG, no período de 2007 a 2016. Realizou-se uma análise exploratória das fichas de notificação do atendimento antirrábico humano do Sistema de Informação de Agravos de Notificações (Sinan). Nos resultados, observou-se que 37,4% das pessoas eram adultas do gênero feminino, com idade entre 20 e 59 anos, 88,7% residentes em área urbana, 26,1% de etnia branca e apenas 31,7% dos pacientes buscaram atendimento antirrábico com até 24 horas da exposição ao vírus rábico. Lesões por mordedura ocorreram em 75,6% dos casos notificados. Quanto às características dos ferimentos, 59,6% foram superficiais, 62,2% ocorreram em mãos/pés e 50,1% foram lesões únicas. Na faixa etária de zero a 10 anos, as regiões anatômicas mãos/pés e cabeça/pescoço/face totalizaram 58,35%. Ressalta-se a necessidade de adoção de ações socioeducativas junto à população, devido ao risco de transmissão de raiva pelo gato, principalmente em áreas em que o vírus rábico circula em populações de morcegos. Sugerem-se melhorias no preenchimento das fichas de notificação do Sinan para melhor compreender o perfil epidemiológico dos pacientes que buscam o atendimento antirrábico e, assim, tornar mais eficaz a gestão desse serviço público.(AU)


The objective of this study was to know the epidemiological profile of the patient who sought post-exposure anti-rabies treatment associated with accidents with domestic cats in Belo Horizonte, Minas Gerais Brazil, from 2007 to 2016. An exploratory analysis of the reports on human rabies of the Notification of Injury Information System was performed. In the results, it was observed that 37.4% of the individuals were female adults aged 20-59 years, 88.7% were urban residents, 26.1% were white, and only 31.7% sought anti-rabies care within 24 hours of exposure to the rabies virus. Bite lesions occurred in 75.6%. Regarding the type of wound, 59.6% were superficial; as for the anatomical location, 62.2% occurred in hands / feet and 50.1% were single lesions. It is necessary to adopt socio-educational actions among the population, due to the risk of transmission of the rabies by the cat, especially in areas where the rabies virus circulates in bat populations. Improvements in the completion of the SINAN notification sheets are suggested to better understand the epidemiological profile of patients seeking anti-rabies treatment and thus improve the management of this public service.(AU)

ABSTRACT

We analyzed a large number of immune response parameters from quarter milk samples with distinct bacteriological and quarter somatic cell count (qSCC) statuses. Furthermore, we sought to explore and identify displayed immune response patterns in milk samples from mammary glands with nonspecific mastitis. Thus, 92 quarter milk samples from 28 cows were stratified into 4 groups, as follows: (1) 49 culture-negative control quarters with a low qSCC (<1 × 105 cells/mL) from 19 dairy cows (so-called healthy quarters); (2) 15 culture-negative quarters with high qSCC (>2 × 105 cells/mL; so-called quarters with nonspecific mastitis) from 10 dairy cows; (3) 8 culture-positive quarters with low qSCC (noninflammatory quarters with low qSCC) from 5 dairy cows; and (4) 20 culture-positive quarters with high qSCC (so-called truly infected quarters) from 8 dairy cows. Using flow cytometry, we evaluated the percentage of milk neutrophils and their viability, intracellular reactive oxygen species production, phagocytosis, and the expression of CD62L, CD11b, and CD44 for each of the 4 quarter strata. Furthermore, the percentage of monocyte/macrophages, B cells, and T lymphocyte subsets were evaluated by flow cytometry. Milk samples from bacteriologically negative quarters (both with a low and elevated qSCC) had a lower qSCC than those with bacteriologically positive outcomes (both with a low and elevated qSCC). As expected, the healthy quarters showed the lowest percentage of neutrophils and also showed a higher percentage of milk monocytes/macrophages and lower percentage of T lymphocytes than truly infected quarters. The most prominent result of the present study is that quarters with nonspecific mastitis showed the highest percentage of milk CD4+ T lymphocytes. The healthy quarters had a lower percentage of apoptotic neutrophils than noninflammatory and truly infected quarters, although it did not differ from those from the quarters with nonspecific mastitis. Our study supports the role of differential cell counting in the diagnosis of mastitis, as the milk leukocyte populations markedly fluctuate under healthy and inflammatory conditions. Furthermore, an increase in milk CD4+ T cells was associated with nonspecific mastitis, suggesting an increase in this leukocyte subpopulation is correlated with low bacterial shedding. Our study allows us to go further in our understanding of mammary gland immunity, providing further insights on potential protective mammary gland immunity, which we hypothesize can open new avenues for the development of novel targets that can promote bovine udder health.

ABSTRACT

The wild cycle of rabies constitutes a serious challenge to epidemiological surveillance for disease control in domestic, companion or production animals, and in humans. The understanding of rabies virus circulation in the natural environment is increasingly important due to the constancy of natural reservoirs of the disease and the presence of potential vectors of the infection to humans and domestic animals. Aiming to evaluate the occurrence of rabies in the State of Sergipe a total of 935 hematophagous bats (Desmodus rotundus), 46 wild dogs (Cerdocyon thous) and 24 primates (Callithrix spp.) were analyzed from 1987 to 2014, of which 1 bat, 17 crab-eating foxes and no primates were positive. Due to the lack of positive results in hematophagous bats, the main vector of herbivorous rabies, more studies are needed to monitor cases, because from an epidemiological point of view, Sergipe is endemic for herbivorous rabies. Epidemiological surveillance of rabies virus in wild animals is primordial for the success of disease control programs in herds of domestic animals and humans.(AU)


O ciclo silvestre da raiva constitui um sério desafio para a vigilância epidemiológica no controle da doença nos animais domésticos, de companhia ou de produção, e nos seres humanos. O entendimento sobre a circulação do vírus rábico no ambiente natural é cada vez mais importante, em razão da constância de reservatórios naturais da doença e da presença de vetores potenciais da infecção aos humanos e aos animais domésticos. Com o objetivo de avaliar a ocorrência da raiva no estado de Sergipe, foram analisados 935 morcegos hematófagos (Desmodus rotundus), 46 cachorros-do-mato (Cerdocyon thous) e 24 primatas (Callithrix spp.) no período de 1987 a 2014, dos quais resultaram positivos um morcego, 17 cachorros-do-mato e nenhum primata. Em que pese a contundente falta de resultados positivos em morcegos hematófagos, principal vetor da raiva dos herbívoros, mais estudos são necessários no monitoramento dos casos, pois o estado, do ponto de vista epidemiológico, é endêmico para a raiva dos herbívoros. A vigilância epidemiológica do vírus da raiva nos animais silvestres é primordial para o sucesso dos programas de controle da doença em rebanhos de animais domésticos e em seres humanos.(AU)

ABSTRACT

O estudo da profilaxia da raiva humana contribui para orientação quanto ao uso de vacinas e soros antirrábicos, servindo de base para a definição de estratégias de prevenção, controle e avaliação da doença. Objetivou-se avaliar o atendimento antirrábico humano pós-exposição e possíveis incompatibilidades com o protocolo do Ministério da Saúde, em agressões por cães, no município de Belo Horizonte. Analisaram-se as fichas de atendimento antirrábico do SINAN para comparação dos tratamentos prescritos com as Normas de Profilaxia antirrábica do Ministério da Saúde. As características dos atendimentos foram: quanto à exposição, 93,0% decorrentes de mordedura; quanto à localização da agressão, 35,5% ocorreram nos membros inferiores; quanto ao ferimento, 62,0% foram lesões únicas; quanto ao tipo de ferimento, 65,8% foram superficiais; quanto à condição do animal, 85,0% ocorreram por animais sadios; em 83,4% dos casos os animais eram observáveis; quanto ao tratamento, observação e vacina (57,3%). O tratamento foi inadequado em 32,7% das condutas. O número de tratamentos instituídos foi muito elevado, 75,7% dos casos. De todos os atendimentos 21,2% das indicações foram consideradas excessivas e 11,5% insuficientes. O sistema de vigilância apresenta falhas e existe a necessidade de corrigi-las para que as informações referentes à finalização dos casos sejam conclusivas.(AU)


The study of prophylaxis of human rabies contributes to guidance on the use of vaccines and anti-rabies serums, providing the basis for the definition of strategies for prevention, control and evaluation of the disease. This study aimed to evaluate the service to early post-exposure human anti-rabies and possible incompatibilities with the Ministry of Health protocol attacks by dogs in the city of Belo Horizonte. The anti-rabies care records of SINAN were analyzed for comparison of treatments prescribed in the Prophylaxis of Rabies Standards of the Ministry of Health. The characteristics of the calls were about the exposure, 93.0% resulting from biting; the location of the assault, 35.5% occurred in the lower limbs; the injury, only 62.0% were injuries; the type of injury, 65.8% were superficial; the condition of the animal, 85.0% occurred in healthy animals; 83.4% of the cases the animals were observed; as treatment, observation and vaccine (57.3%). The treatment was inadequate in 32.7%. The number of established treatments was very high, 75.7% of cases. In all 21.2% of indications consultations were considered excessive and 11.5% insufficient. The surveillance system is faulty and there is a need to correct them so that the information regarding the completion of cases are conclusive.(AU)

ABSTRACT

This study identified potential blood markers associated with mastitis in dairy cows under different dry therapies during the transition period, using a logistic regression model. Thirty-four Holstein dairy cows were divided into three groups: untreated controls (13 cows, 42 quarters); animals that received an antimicrobial at drying-off (10 cows, 40 quarters); and animals that were administered an internal teat sealant at drying-off (11 cow, 44 quarters). Blood and quarter milk samples were collected 60 days before the expected day of calving, on the day of calving, and three, seven, 15, 21, and 30 days after calving. Milk samples were submitted for bacteriological analysis and somatic cell count. Blood samples were collected for analyses of the following: the erythrogram and leucogram; plasm fibrinogen concentration; hepatic and renal functions; metabolic profile; serum calcium and phosphorous levels; total serum protein and albumin concentrations. The concentration of total serum proteins was associated with a high somatic cell count. Similarly, the concentrations of total serum proteins and triglycerides were associated to milk bacteriological positive samples during the transition period. Thus, the occurrence of mastitis in dairy cows during the transition period was greater in animals that showed higher concentrations of serum total proteins and triglycerides, in contrast to the use of dry cow therapy.(AU)


O presente estudo identificou potenciais marcadores sanguíneos associados tanto à mastite, durante o período de transição em vacas leiteiras submetidas à antimicrobianoterapia, quanto à utilização do selante interno de teto na secagem por modelo de regressão logística. Trinta e quatro vacas da raça Holandesa foram divididas em três grupos experimentais, a saber: 13 animais (42 quartos mamários) que não receberam tratamento na secagem; 10 animais (40 quartos mamários) tratados por via intramamária com uma bisnaga do antimicrobiano para vaca seca à base de cefalônio anidro após a última ordenha em cada quarto mamário; e 11 animais (44 quartos mamários) que receberam, por via intramamária, uma bisnaga de selante à base de subnitrato de bismuto após a última ordenha em cada quarto mamário. As amostras de leite e sangue foram coletadas 60 dias antes da data prevista do parto, na secagem, e no dia do parto e após três, sete, 15 e 21 dias após o parto. As amostras de leite foram utilizadas para o exame microbiológico e a determinação da contagem de células somáticas. As amostras de sangue foram utilizadas para determinação do eritrograma, leucograma, fibrinogênio plasmático, funções hepáticas e renais, perfil metabólico, proteína total e albuminas séricas e concentração sérica de cálcio e fósforo. A concentração total de proteínas séricas foi associada à alta contagem de células somáticas. Similarmente, a concentração total de proteínas séricas e triglicérides foi associada ao isolamento de patógenos causadores de mastite nas amostras de leite durante o período de transição. Desse modo, conclui-se que vacas leiteiras com concentrações séricas maiores de proteína total e triglicérides têm maior chance de apresentar mastite durante o período de transição; em contraste, o uso da antimicrobianoterapia de vaca seca reduz esse risco.(AU)

ABSTRACT

O presente estudo objetivou avaliar o perfil das Ig durante os diferentes tratamentos de vacas com mastite clínica. Para isso, 30 vacas com mastite clínica em um quarto mamário foram utilizadas e divididas em três grupos. O primeiro grupo foi composto por 10 animais submetidos ao tratamento combinado com infusão intramamária de 8,5mg de sulfato de cefquinoma após cada ordenha, totalizando três aplicações e administração intramuscular de 2,5mg/kg de enrofloxacina por três dias. O segundo grupo foi composto por 10 animais submetidos ao tratamento intramamário, com infusão intramamária de 8,5mg de sulfato de cefquinoma, após cada ordenha, totalizando três aplicações. O terceiro grupo foi composto por 10 animais submetidos ao tratamento sistêmico, com 2,5mg/kg de enrofloxacina, durante três dias. As amostras de leite foram coletadas de todos os animais antes dos tratamentos (momento 0), no segundo (momento 1), no quinto (momento 2) e no 12º dia (momento 3) após o término dos tratamentos. Estas foram submetidas à contagem de células somáticas, ao California Mastitis Test (CMT), ao exame bacteriológico e à quantificação das IgG1, IgG2, IgA e IgM. O tratamento combinado foi mais eficaz e precoce na taxa de cura clínica, na redução dos escores de CMT e da contagem de células somáticas. Além disso, os resultados do presente estudo demonstraram que as concentrações lácteas das diferentes classes de Ig, apesar de sua importância biológica, não estão relacionadas ao prognóstico da mastite clínica bovina, ou seja, não podem ser consideradas marcadores robustos associados à cura clínica e/ou bacteriológica da infecção intramamária.(AU)

Here, 30 Holstein cows with clinical mastitis in one quarter were uniformly divided into: 10 dairy cows that received 8.5mg of cefquinome sulphate administrated intramammarily during three consecutive milkings and 2.5mg/kg of enrofloxacin administrated parenterally during three consecutive days (Group 1); 10 dairy cows that received 8.5mg of cefquinome sulphate administrated intramammarily during three consecutive milkings (Group 2); and 10 dairy cows that received 2.5mg/kg of enrofloxacin administrated parenterally during three consecutive days (Group 3). Milk samples for somatic cell count, California Mastitis Test (CMT), bacteriological culture and quantification of IgG1, IgG2, IgM and IgA were collected before antimicrobial treatment, and after two, five and 12 days after the antimicrobial treatment. The combined treatment was more effective in the clinical cure rate, reduction of somatic cell count and CMT scores. Furthermore, the results demonstrated that milk concentration of different Igs classes were not related to prognosis of bovine clinical mastitis, and then cannot be considered as robust markers associated with clinical and bacteriological cures.(AU)

ABSTRACT

709 clinical mastitis cases were analyzed and treated with antimicrobial combination cephalexin-neomycin and the anti-inflammatory prednisolone. A sample of milk was collected to perform a microbiological culture before starting the treatment and 14 days later. Somatic cell count (SCC) was obtained from samples collected on the day of the clinical case (D0), 14 days after (D14) and 28 days after (D28). Of the total, 435 (61.4%) at the D0 exhibited growth of microorganisms. Of the isolated agents, 365 (84%) were Gram-positive, and 66 (16%) were Gram-negative. A clinical cure was achieved in 63% of cases. Bacteriological cure occurred in 75% of cases. Only at D28 after the clinical case a significant SCC reduction was verified. The logistic regression for clinical cure showed significant effects for days in milk and parity (P< 0.05). For bacteriological cure, there were significant effects of Log (SCC) D0; clinical cure and quarter affected (P< 0.05). In the principal component analysis, the Temperature-Humidity Index was associated with reduced clinical cure of clinical mastitis cases.(AU)


Setecentos e nove casos clínicos de mastite foram analisados e tratados com combinação antimicrobiana à base de cefalexina-neomicina e o anti-inflamatório prednisolona. Uma amostra de leite foi coletada para realização de cultura microbiológica antes do início do tratamento e 14 dias depois. A contagem de células somáticas (SCC) foi obtida de amostras coletadas no dia do caso clínico (D0), 14 dias após (D14) e 28 dias após (D28). Do total, 435 (61,4%) no D0 apresentaram crescimento de microrganismos, enquanto em 274 (38,6%) não houve crescimento. Dos agentes isolados, 365 (84%) eram Gram-positivos e 66 (16%) eram Gram-negativos. A cura clínica foi alcançada em 63% dos casos. A cura bacteriológica ocorreu em 75% dos casos. Apenas no D28 verificou-se uma redução significativa na SCC. A regressão logística para a cura clínica mostrou efeitos significativos para dias em lactação e paridade (P<0,05). Para a cura bacteriológica, houve efeitos significativos de Log (SCC) D0; cura clínica e quarto afetado (P<0,05). Na análise do componente principal, o índice de temperatura-umidade foi associado com a redução da cura clínica dos casos clínicos da mastite.(AU)

The present study attempted to identify individual milk proteins and other milk components that are associated with casein micelle size (CMS) and dry matter cheese yield (DMCY) using factor analysis. Here, we used 140 bulk tank milk samples from different farms. Milk composition was determined using a Fourier transform infrared equipament. The individual milk proteins were (αS-casein, ß-casein, κ-casein, ß-lactoglobulin and α-lactoalbumin) measured by their electrophoretic profile. The CMS was estimated by photon correlation spectroscopy, and the DMCY was determined using reduced laboratory-scale cheese production. Factor analysis partitioned the milk components into three groups that, taken together, explain 68.3% of the total variance. The first factor was defined as "CMS", while the second as "DMCY" factor, based on their high loadings. The CMS was positively correlated with protein, casein, non-fat solids and αS-casein and negatively associated with κ-casein and ß-lactoglubulin. DMCY was positively correlated with fat, protein, casein, total solids and negatively correlated with αs-casein. These results indicate that the variation of individual milk proteins may be an important aspect correlated to milk quality and cheese production.(AU)


O objetivo do presente estudo foi avaliar a associação das frações proteicas individuais e de outros componentes do leite com o tamanho das micelas de caseína (TMC) e a produção de matéria seca de queijo (MSQ) utilizando-se análise fatorial. Foram coletadas 140 amostras de leite de tanque provenientes de diferentes fazendas. A determinação da composição do leite foi determinada por espectroscopia no infravermelho com transformação de Fourier. As proteínas individuais (αS-caseína, ß-caseína, κ-caseína, ß-lactoglobulina e α-lactalbumina) foram quantificadas pelo perfil eletroforético. O tamanho médio das micelas de caseína foi analisado pelo princípio de espectroscopia de correlação de fótons e pela produção MSQ a partir do modelo de coagulação do leite em escala reduzida. A análise fatorial delimitou as variáveis em três fatores, que, juntos, responderam por 68,3% da variação total dos dados. No primeiro fator foram observadas as associações mais fortes com o TMC, enquanto no segundo fator as correlações foram mais significativas com a MSQ. O TMC foi associado positivamente com o conteúdo de proteína, caseína, sólidos desengordurados e αS-caseína, e negativamente com κ-caseína e ß-lactoglubulina. MSQ foi associada positivamente com o teor gordura, proteína e caseína total, sólidos totais, e negativamente com o teor de αs-caseína. Esses resultados indicam que a variação quantitativa das proteínas do leite pode ser determinante da qualidade do leite na produção de queijo.(AU)

Abstract Bacanga River Basin faces environmental problems related to urbanization and discharge of untreated domestic sewage, which compromise its ecosystem health. Due to the small number of studies that assessed its water quality, the present study aimed to assess the current status of this ecosystem based on the aquatic life protection index. Samples were carried out every two months, in a total of six events, in six sites along the basin, where the water samples were collected to assess physicochemical parameters and calculate the trophic state index and the index of minimum parameters for the protection of aquatic communities. The data were also compared with values determined by the resolution National Environment Council - CONAMA 357/05. Our results reveal significant changes in the water quality of Bacanga River Basin. An increase in nutrients and chlorophyll-a concentration led it to eutrophication. The surfactant values were high and put in danger the aquatic biota. Dissolved oxygen rates were below the values allowed by the resolution in most sites sampled. The current water quality is terrible for the protection of aquatic life in 61.92% of the sites sampled.


Resumo A Bacia Hidrográfica do rio Bacanga (BHRB) apresenta problemas ambientais relacionados a urbanização e lançamentos de esgoto in natura que comprometem a qualidade desse ecossistema. Devido ao reduzido número de estudos associados à avaliação da qualidade da água no local, este estudo teve como objetivo avaliar a situação atual desse ecossistema por meio do Índice de Proteção da Vida Aquática. Seis amostragens bimestrais foram realizadas em seis pontos ao longo da bacia, coletando parâmetros físico-químicos para a aplicação do Índice de Estado Trófico e Índice de Parâmetros Mínimos para a Proteção da Vida Aquática, relacionando-os com a resolução Conselho Nacional do Meio Ambiente - CONAMA/357. Os resultados revelaram alterações significativas na qualidade da água da BHRB, o aumento de nutrientes e das concentrações de clorofila-a conduziram ao um estado geral de eutrofização. Os valores de surfactantes foram altos colocando em risco a biota aquática e as taxas de oxigênio dissolvido estiveram abaixo do permitido pela resolução na maioria dos pontos amostrados. A situação atual da qualidade da água para proteção da vida aquática é péssima em 61,92% dos pontos amostrados.

ABSTRACT

Bovine digital dermatitis (BDD) is an infectious and contagious disease characterized by ulcerative and proliferative lesions affecting the skin on the bulbs of the heel or the interdigital cleft in dairy cattle, often associated with lameness. Evidences on the etiology of BDD indicate that it is multifactorial, involving environmental factors and multiple bacterial colonization. We isolated and identified microorganisms from BDD biopsy samples obtained from five Holstein Friesian and two Jersey cows by cultivation and molecular identification of bacterial isolates using 16S rRNA gene sequence analysis. We identified six bacterial species: Spirochetes as Treponema pedis and Leptospira broomi/L. fainei, L. licerasiae/L. wolffii; Corynebacterium appendicis, Cupriavidus gilardii and Enterococcus casseliflavus/E. gallinarum. It was quite surprising to have isolated and identified Leptospira species in three out of seven cultures, from different individual cows and two different farms. The species identified belong to the intermediate pathogenic clade, which is a group found to cause human and animal disease. Our findings indicate the need to further investigate the association of Leptospira of intermediate pathogenicity with BDD lesions and whether its presence would have any veterinary and medical significance both in Leptospirosis and with the pathogenesis of BDD lesions, especially in tropical countries.(AU)


Dermatite digital bovina (DDB) é uma doença infecciosa, contagiosa, caracterizada por lesões ulcerativas e proliferativas da região dos talões e/ou do espaço interdigital, frequentemente associada com claudicação. Evidências indicam que a etiologia da DDB é multifatorial, envolvendo fatores ambientais e colonização polimicrobiana. Relata-se aqui o isolamento e a identificação bacteriana em amostras de biópsias em lesões de DDB, obtidas de cinco vacas da raça Holandesa e duas da raça Jersey, por meio de cultivo e identificação molecular de isolados, com base na análise de sequências de genes 16S rRNA. São identificadas seis espécies bacterianas: as espiroquetas Treponema pedis e Leptospira broomi/L. fainei, L. licerasiae/L. wolffii; Corynebacterium appendicis, Cupriavidus gilardii e Enterococcus casseliflavus/E. gallinarum. O isolamento e a identificação de espécies de Leptospira surpreenderam, destacando-se sua presença em três dos sete cultivos obtidos em diferentes vacas, de duas fazendas distintas. As espécies identificadas pertencem ao grupo tipificado como de patogenicidade intermediária, causador de doenças em animais e no homem. Os resultados apresentados indicam a necessidade de maiores investigações sobre a associação entre Leptospira de patogenicidade intermediária e a patogênese das lesões DDB, investigando-se sua presença e significado nas medicinas veterinária e humana, especialmente em países tropicais.(AU)

ABSTRACT

This study examined neutrophil and monocyte functions and the blood lymphocyte profile of naturally BLV-infected cows with or without persistent lymphocytosis (PL). The percentage of neutrophils and monocytes that phagocytosed Staphylococcus aureus was lower in BLV-infected dairy cows, particularly those with PL. The relative percentage of CD44+ monocytes and neutrophils and CD11b expression by neutrophils was also lower in BLV-infected dairy cows with PL. A correlation between the percentage of CD11b+ neutrophils and that produced reactive oxygen species (ROS) was found. Furthermore, the percentage of CD44+ monocytes was positively correlated with the percentage of monocytes that phagocytosed S. aureus and the same phenomenon was observed for neutrophils. In BLV-infected dairy cows, particularly those with PL, inhibition of monocyte and neutrophil apoptosis was observed. Additionally, the percentage of neutrophils producing ROS was lower in BLV-infected cows with PL, in contrast to higher intensity of intracellular production of ROS by monocytes. The result from the lymphocyte immunophenotyping of BLV-infected cows with PL was an increase in B cells, mainly B CD5+ CD11b+, due to the apoptosis inhibition. In conclusion, this study provides novel insight into the implications of BLV infection for cattle, which can include the dysfunction of blood monocytes and neutrophils.

ABSTRACT

The present study aimed to identify the climate condition parameters that are associated with the occurrence of antimicrobial and macrocyclic lactone residues in bulk tank milk using a multivariate principal components analysis (PCA). A total of 132 raw milk samples were collected at dairy farms in Minas Gerais State in Brazil and analyzed for 35 analytes, comprising macrocyclic lactones and antibacterials, using liquid chromatography coupled with mass spectrometry in tandem mode spectrometry. Of the 132 samples, 34 (25.76%) bulk tank milk samples were positive for at least one analyte. PCA showed that antimicrobial residues in bulk tank milk occurred less frequently on days with a higher average temperature, maximum temperature and temperature-humidity index. In contrast, relative humidity was inversely associated with antimicrobial residues in raw milk. The PCA showed that daily milk production was also related to macrocyclic lactone residues, while rainfall showed an inverse association. Thus, some climate conditions, such as average temperature, maximum temperature and temperature-humidity index, can predict the moments with lower risk of occurrence of antimicrobial residues in bulk tank milk, in contrast to relative humidity. Furthermore, the risk of macrocyclic lactone residues in bulk tank milk was higher in months with less rainfall.(AU)


O presente trabalho objetivou identificar fatores climáticos associados à ocorrência de resíduos de antimicrobianos e lactonas macrocíclicas em amostras de leite de tanque por análise multivariada de componentes principais (ACP). Para o presente trabalho, 132 amostras de leite cru foram coletadas em fazendas leiteiras localizadas no estado de Minas Gerais (Brasil) e analisadas por cromatografia líquida de alta eficiência e espectrometria de massas in tandem para detecção de 35 analitos, incluindo antimicrobianos e lactonas macrocíclicas. Das 132 amostras de leite analisadas, detectou-se pelo menos um analito em 34 (25,76%) amostras. A ACP demonstrou que a presença de resíduos de antimicrobianos no leite de tanque ocorreu menos frequentemente nos dias com maior temperatura média, temperatura máxima e índice de temperatura e umidade. Por outro lado, a umidade relativa foi inversamente associada à presença de resíduos antimicrobianos no leite. A ACP demonstrou associação entre a presença de resíduos de lactonas macrocíclicas no leite e a produção diária de leite, e a presença de resíduos de lactonas macrocíclicas ocorreu menos frequentemente nos meses com maiores índices pluviométricos. Dessa forma, conclui-se que alguns índices climáticos, como temperatura média, temperatura máxima e índice de temperatura e umidade, podem predizer períodos com maior risco de ocorrência de resíduos de antimicrobianos, em contraste com a umidade relativa. Além disso, o risco de resíduos de lactonas macrocíclicas no leite de tanque foi maior nos meses com menores índices pluviométricos.(AU)

The present study attempted to identify climate conditions that are associated with the occurrence of pyrethroid residues using a multivariate principal component analysis (PCA). A total of 132 raw milk samples were collected from dairy farms in Minas Gerais State - Brazil and analyzed for seven analytes using gas chromatography with electron capture detection. Of the 132 milk samples, pyrethroid residues were identified in 14 (10.6%) milk samples, of which 12 (9.1%) and two (1.5%) milk samples had the identification of cypermethrin and deltamethrin, respectively. From those samples, nine (6.8%) milk samples were regarded as non-compliant for cypermethrin with this analyte concentration above the maximum residue limits set by Brazilian legislation. A PCA assessing pyrethroid residues in bulk tank milk demonstrated that the average temperature and the Temperature-Humidity Index were associated with pyrethroids residues in bulk milk tank, although the relative humidity was inversely correlated. Thus, the data analysis indicated that the pyrethrid residues associated with some climate conditions can predict the moments with higher risk of occurrence of pyrethroid residues in bulk tank milk.(AU)


O presente trabalho objetivou identificar fatores climáticos associados à ocorrência de resíduos de piretroides em amostras de leite de tanque por análise multivariada de componentes principais (ACP). Para o presente trabalho, 132 amostras de leite cru foram coletadas em fazendas leiteiras localizadas no estado de Minas Gerais (Brasil) e analisadas por sete analitos por cromatografia gasosa com detector por captura de eletróns para detecção de resíduos de piretroides. Das 132 amostras de leite analisadas, a presença de resíduos de piretroides foi detectada em 14 amostras (10,6%), e em 12 (9,1%) e duas (1,5%) foram detectados cipermetrina e deltametrina, respectivamente. Destas amostras, em nove (6,8%) a concentração de cipermetrina encontrada foi maior que a permitida pela legislação brasileira. A ACP demonstrou que a presença de resíduos de piretroides no leite de tanque ocorreu menos frequentemente nos dias com maior umidade relativa, embora a presença desses resíduos estivesse associada a maior temperatura média e a maior índice de temperatura e umidade. Dessa forma, conclui-se que alguns índices climáticos podem predizer períodos com maior risco de ocorrência de resíduos de piretroides no leite de tanque.(AU)

ABSTRACT

Após validação de dois diferentes métodos de mensuração de pressão arterial sistólica (PAS) não invasivos ou indiretos (oscilométrico e Doppler vascular) com o invasivo ou direto (cateterização arterial - padrão ouro) em 12 gatos hígidos anestesiados de mesma faixa etária, os métodos não invasivos foram avaliados e comparados em 24 gatos hígidos e conscientes, divididos em quatro grupos de idade. Em cada animal, procedeu-se à mensuração da pressão pelos métodos Doppler e oscilométrico. O método oscilométrico foi o primeiro a ser realizado, e, logo em seguida, o método Doppler. Os valores considerados foram obtidos das médias de sete medidas consecutivas para cada método, com intervalo de 30 segundos entre elas. A primeira medida e as medidas discrepantes (variação ˃ 20% PAS), obtidas com sinais óbvios de estresse e/ou com movimentação do animal, foram descartadas. Os métodos de mensuração oscilométrico e Doppler, embora tenham apresentado médias estatisticamente diferentes em todos os grupos, quando comparados em cada grupo, apresentaram uma correlação significativa, alta e positiva. Ou seja, sempre que for obtido um valor elevado por um método, o mesmo fato ocorrerá com o outro método e vice-versa. Clinicamente, a diferença observada não é relevante, uma vez que a diferença média dos valores obtida pelos dois métodos, em cada grupo, foi menor que 1,1%.(AU)


After two different validation systolic blood pressure measurement methods (PAS), not invasive or indirect (oscillometric and doppler vascular) with invasive or direct (arterial catheterization - Gold Standard) in 12 anesthetized healthy cats of the same age group, not invasive methods were evaluated and compared in 24 healthy conscious cats, divided into four age groups. In each animal, we proceeded to measure the pressure by Doppler and oscillometric methods. The oscillometric method was the first to be performed, and, shortly thereafter, the doppler method. The values considered were obtained from averages from seven consecutive measurements for each method with an interval of thirty seconds between them. The first measurement and disparate measurements (range ˃ 20% PAS) obtained with obvious signs of stress and / or animal movement were discarded. Methods of oscillometric and doppler measurement, although statistically different averages were shown in all groups when compared, each group showed a significant high positive correlation, meaning that when a high value is obtained by a method, the same occurs with the other method, and vice versa. Clinically, the observed difference is not relevant since the mean difference values obtained by the two methods, in each group was lower than 1.1%.(AU)
